Monitoring of antioxidant properties of uric acid in humans for a
consideration measuring of levels of allantoin in plasma by liquid
chromatography.

Uric acid is the end product of purine metabolism in humans. It has
been pointed out that uric acid acts as an antioxidant and is capable
to react with biologically relevant oxidants to form allantoin.
Therefore, measurement of allantoin in humans was proposed as a marker
of oxidative stress. We estimated allantoin in human plasma obtained
from the patients with chronic renal failure before hemodialysis
(n=30), patients with non-insulin dependent diabetes mellitus (n=30)
and blood donors (n=30) using a method based on selective isolation of
allantoin from deproteinized plasma samples on AG 1-X8 resin and its
derivatization to glyoxylate-2,4-dinitrophenylhydrazone. The method is
free from urate and glyoxylate interferences. Separation of
glyoxylate-2,4-dinitrophenylhydrazone from other hydrazones was
achieved on reversed phase HPLC with gradient elution and UV/VIS
detection at 360 nm. The analytical performance of this method is
satisfactory with intra-assay CV 5.7%, inter-assay CV 8.3% and recovery
94.1%. We have determined other parameters of oxidative stress
(malondialdehyde, total antioxidant status, superoxide dismutase and
glutathione peroxidase) too. The preliminary reference range of
allantoin in a group of blood donors is 4.76+/-2.99 mumol/L. In the
patients with chronic renal failure and the patients with non-insulin
dependent diabetes mellitus we found allantoin levels in plasma
(27.1+/-13.8) mumol/L and (11.08+/-5.90) mumol/L, respectively. It
seems that allantoin is a possible indicator of free radical damage in
vivo.

The recent finding of the beneficial effects of iron deprivation in the
outcome of muscle necrosis in an animal model of genetic myopathy
served as the basis of this commentary. Here, "taking away" iron by
controlled dietary deprivation is proposed as a reasonable, feasible,
cheap, and efficient clinical approach to many diverse diseases, all of
which have a free radical component. Indeed, iron potentiates the
generation of the highly reactive and toxic hydroxyl radical, and,
thus, of oxidative damage. Iron deprivation may represent the first
really efficient antioxidant, preventing oxidative stress in all
subcellular compartments, tissues, and organs. Iron/iron deprivation
also modulates programmed cell death (apoptosis), which should be the
subject of further studies to better define the mechanisms mediating
these complex effects. Finally, related to its antioxidant effects,
iron deprivation may find applications in the anti-aging field, whether
programmed or premature aging, and whether in cosmetics or in

conclusion, therapy by taking away (iron) has a great potential formany
different diseases, all of which share ROS-mediated mechanisms.
Thedevelopment of new, non-toxic , easily administrable iron chelators
suchas IRCO11 may shortly become the most efficient and
fashionableantioxidant, anti-aging, anti-infectious, and
anti-inflammatroy therapy.In the meantime, although taking away by
controlled dietary deprivation isless attractive , it should be
considered in all of the above, as well asin the currently incurable,
devastating genetic or acquired myopathiessuch as DMD.

Didn't you once tell me it was dld mthrfkr dld ?

Anyway - back to the topic - blood donors are used as all manner of
"standard models" as their clearly healthy diet enables them to maintain
healthy levels of various parameters over consectuive donations.
However pathological blood letters just become listless and anaemic, and
suffer from a range of debilitating symptoms.
