Oxidative stress is induced under diabetic conditions through various
pathways, including the electron transport chain in mitochondria and
the nonenzymatic glycosylation reaction, and is likely involved in
progression of pancreatic beta-cell dysfunction developing in diabetes.
beta-Cells are vulnerable to oxidative stress, possibly due to low
levels of antioxidant enzyme expression. When oxidative stress was
induced in vitro in beta cells, the insulin gene promoter activity and
mRNA levels were suppressed, accompanied by the reduced activity of
pancreatic and duodenal homeobox factor-1 (PDX-1) (also known as
IDX-1/STF-1/IPF1), an important transcription factor for the insulin
gene. The suppression of oxidative stress by a potent antioxidant,
N-acetyl-l-cysteine or probucol, led to the recovery of insulin
biosynthesis and PDX-1 expression in nuclei and improved glucose
tolerance in animal models for type 2 diabetes. As a possible cause of
this, we recently found that PDX-1 was translocated from the nucleus to
the cytoplasm in response to oxidative stress. Furthermore, the
addition of a dominant-negative form of c-Jun N-terminal kinase (JNK)
inhibited the oxidative stress-induced PDX-1 translocation, suggesting
an essential role of JNK in mediating the phenomenon. Taken together,
the oxidative stress-mediated activation of the JNK pathway leads to
nucleocytoplasmic translocation of PDX-1 and thus is likely involved in
the progression of beta-cell dysfunction found in diabetes.

"involved" is it, now you are catching on. Will we soon seen an amended
iron causes all disease because people eat meat idea modification? The
world's highest rate of diabetes is found in a population who eat few or
no animal products, so the meat part is defunct. They also often suffer
from low iron, the world's number one health problem, so the "iron causes"
is also defunct. An amended idea will be fine, that's what real science
does when it finds the evidence doesn't support an idea, it is either
tossed or amended.

| No, the "involved" is what iron has to do with the topic, not cause, that
is what you are being forced bit by bit to accept. Once accepted you can
then modify the iron causes all disease becauase people eat meat idea and
make it a qualified idea that it can be "involved" but not cause
disorders. We await your revision to bring it into line with the
evidence, other wise called doing science.

| As you come round to our way of thinking,ie. science, you are better
grasping that "involved" is not "cause" any way it can be spun. Oxygen is
"involved" in more biological processes then is iron and using your logic
it "causes" anything in any abstract in which it is mentioned. Not to
fret, this is hard, giving up a long held dearly cherished idea and having
to modify it to fit the evidence, but that is why you are now moving
closer to science, if in the end obsession doesn't get you.
